Sample Preparation
For indicator organisms (TC, EC, YM, XSA):
For indicator organisms (TC, EC, YM, XSA):
- For Filterable liquid samples – Filter 100 ml of liquid sample using a 47 mm membrance filter with 0.45µm pore size.
- For non filterable liquid and powder samples – Weigh standardized sample size (e.g 10g or 25g). Place in a sterile container, then add the appropriate volume of sterile diluent (e.g 90ml or 225ml of Buffer Peptone water or BPW, Maximum Recovery Diluent or MRD, or 0.1% Peptone Water). Homognize and dilute accordingly.
For Pathogens (SL and LS): Weigh 25g and dilute with 225ml sterile BPW.

Refer to codex Alimentarius, Food & Drug Administration (FDA), Food Standards Agency (FSA), or other local regulatory bodies.
Processing notes
Check sample pH after the 10-1 dilution. Ensure that sample pH is between 6 and 8, otherwise neutralize using sterile 0.1N Sodium Hydroxide (NaOH) if lower than 6, or 0.1N Hydrochloric Acid (HCl) if higher than 8.
For filtered samples, rehydrate CompactDryTM plates with 1mL sterile diluent before transferring membrance filter using a rolling motion to avoid gas pockets between the filter and the fabric.

Like other tests that rely on β-Glucuronidase activity to detect E.Coli, CompactDryTM EC cannot detect E.coli O157:H7 as an E.coli.
